BEST ANSWER: Yes they need a gasket sold separately. Typically the clamp will have the gasket with it.

4 inch Tri clamp Cap with pre-drilled hole

I used this to fabricate a top for my converted-keg fermenter. I attached a 1/2 inch NPT fitting which accepts a Blichmann style quick connect which I use for a blow off hose. The hole was the perfect size.
